无多糖巴戟天基因组DNA提取物的制备
Genomic DNA Extraction from Morinda officinalis How
【摘要】 由于巴戟天(MorindaofficinalisHow)植株中多糖含量较高,故其总DNA的提取明显存在着多糖干扰的问题。多糖可以抑制多种生物学酶类(如聚合酶、连接酶、限制性内切酶等)的活性,对DNA的浓缩也有影响;受多糖污染的DNA呈粘稠的胶状,不便于后续的DNA分子生物学实验操作。笔者在对比多种DNA提取方法的基础上,确定先对材料进行一定的预处理,再用CTAB法提取基因组DNA。这种方法,不但有效地解决了巴戟天基因组DNA受多糖干扰的问题,而且仅需使用常用试剂,实验成本低,简便易行。
【Abstract】 Because of the high levels of polysaccharide in Morinda officinalis How,it is difficult to extract high quality and quantity of genomic DNA from its tissues using the conventional methods.Polysaccharide can restrain the activity of many enzymes such as polymerase,ligase and restriction enzymes.And DNA contaminated by polysaccharide is colloidal,which makes the downstream molecular biologic experiments inconvenient.Genomic DNA was successfully extracted from Morinda officinalis How using the hexadecyltrimethylammonium bromide(CTAB)method.Leaves of Morinda officinalis How were pretreated before extracting genomic DNA.The method is simple and feasible.
