卵巢癌组织中CD44V6和VEGF及nm23-H1细胞因子表达的临床意义
Expression of CD44V6,VEGF and nm23-H1 on Ovarian Cancerous Tissue and Its Clinical Significance
【摘要】 目的 :探讨CD44V6、VEGF及nm2 3 H1在卵巢癌等的表达情况及其临床意义。方法 :选取 6 5例手术切除卵巢癌标本、卵巢良性肿瘤 30例和正常卵巢组织 2 9例 ,常规 10 %甲醛固定 ,石蜡包埋 ,连续切片 ,以针对CD44V6、VEGF和nm2 3 H1单克隆抗体 ,SABC免疫组化染色 ,观察了其在卵巢癌、良性卵巢肿瘤及正常卵巢组织的表达并分析相关的临床病理因素。结果 :1)CD44V6和VEGF两细胞因子在卵巢癌、临床Ⅱ~Ⅳ期、生存期 <5年的患者癌组织内 ,其阳性率分别高于良性瘤和正常卵巢组、临床Ⅰ期、生存期 >5年者 ,P <0 0 5 ;CD44V6在转移癌的阳性率亦显著高于原发灶 ,但VEGF阳性率在原发灶和转移瘤间差别不明显。 2 )nm2 3 H1阳性率在临床Ⅱ~Ⅳ期、癌转移灶、癌患者生存期 <5年的癌组织内 ,分别显著低于临床Ⅰ期、原发癌和生存期 >5年者 ,P <0 0 5 ;nm2 3 H1阳性率在卵巢癌中亦显著高于良性瘤、正常卵巢组。 3)未看出 3种因子表达在卵巢癌患者不同年龄组、不同组织类型、病理分级及残留灶大小间其阳性率有明显差异。结论 :1)CD44V6和VEGF表达在分化差的肿瘤组织中较高 ,且患者预后差。 2 )nm2 3 H1在肿瘤早期、原发癌表达明显高于晚期转移癌 ,但其在恶性瘤比良性瘤、正常卵巢组织中阳性率高的结果 ,可能是机体遏制肿瘤、
【Abstract】 Objective To detect the expression of CD 44V 6, VEGF and nm23-H1 genes and evaluate its clinical significances on ovarian cancerous tissue.Methods Monoclonal antibodies to CD 44V 6,VEGF,nm23-H1 and SABC immunohistochemistry method were used to detect the expression of CD 44V 6,VEGF and nm23-H1 on the tumor cell in 65 cases of human ovarian cancerous tissue,30 benign ovarian tumor and on the normal ovarian tissues.Results 1) The positive expression rates of both CD 44V 6 and VEGF in groups of ovarian cancers, of stages Ⅱ-Ⅳand of the patient’s survival below 5 years were respectively were higher than that in groups of benign tumors and normal ovarian controls,of stage Ⅰand of the patient’s survival above 5 years,P<0.05.The expression rate of CD 44V 6 in metastatic lesions were higher than that in the primary tumor,but that of VEGF was not significant between them.2) The positive expression rate of nm23-H1 in groups of stage Ⅱ-Ⅳ,of the metastases and of the patient’s survival below 5 years were respectively lower than that in the groups of stage Ⅰ-Ⅱ,of primary lesions and of the patient’s survival above 5 years,P<0.05.The expression rate of nm23-H1 gene in group of ovarian cancers was higher than that of benign tumor or normal controls too.3) No significant difference was abserved between groups of the patient’s age,of histological types of cancers,of pathological grades of the malignancy and sizes of residual focuses of cancer.Conclusions 1) The results in the experiment indicate that as the expression of CD 44 V6 and VEGF is higher in poor differentiated tmuor and when the diseases reach the lase stage,which is concordance with some documents in which CD 44V 6 is an agent promoting growth, infiltration and metastasis of tumor,and VEGF promote tumor’s vessel growth.2) The higher expression of nm23-H1 primary cancer than that on metastatic lesion in late stage also reflect the function of nm23-H1 as metastasis supressor gene. The result of higher expression of nm23-H1 in cancer group than that in benign tumors or normal controls may be considered as a host compensative phenomenon to suppress tumor.
