重组人白细胞介素-6治疗小鼠膀胱癌机制的实验研究

【摘要】 目的 :探讨重组人白细胞介素 - 6 (rhIL 6 )对BTT739荷瘤小鼠的抗肿瘤作用机制。方法 :BTT739肿瘤细胞接种T739小鼠 ,随机分组 :1)对照组 :腹腔注射生理盐水 0 2mL ,每日 1次 ;2 )rhIL 6组 :每日腹腔注射 1次rhIL 6 ,剂量为 4× 10 6IU/kg小鼠体重 ;应用电镜、流式细胞分析技术检测细胞凋亡 ;用流式细胞仪检测肿瘤细胞膜Fas与FasL受体及Bcl 2蛋白的表达。结果 :rhIL 6组小鼠肿瘤组织电镜下可见特征性凋亡细胞的出现 ,流式细胞仪检测出在G1期前出现凋亡峰较对照组明显增加 ,P <0 0 5 ;Fas与FasL的表达较对照组明显增加 ,rhIL 6组升至 12 5 7%和 2 0 1% ,而对照组仅为 4 6 6 %和 14 1% ,经检验P <0 0 5 ;Bcl 2蛋白的表达较对照组无明显变化 ,经检验P >0 0 5。结论 :rhIL 6可以诱导小鼠膀胱癌细胞凋亡 ,死亡受体Fas及FasL的表达增加 ,可能是rhIL 6诱导肿瘤细胞凋亡的重要信息传导途径
【Abstract】 Objective To investigate the mechanism of antitumor with the rhIL-6 on the mouse bladder carcinoma BTT739. Methods After inoculated with the bladder tumor BTT739 cells,the mice were randomly divided into 2 groups:1)Group A as a control group;2)Group B was given abdominal cavity injection of rhIL-6 in a dosage of 4×10 6 IU/kg per day;the electronmicroscopy,flow cytometry techniques were used to test the expression of Fas and FasL receptor and Bcl-2 protein,and its morphological changes of apoptosis. Results Compared with the control, the tumor cells of the rhIL-6 group showed a typical apoptotic morphology and hypodiploid peak before G 1 phase,increased the expression of Fas and FasL of the tumor cells,from 4.66%,14.1% to 12.57% and 20.1%,P<0.05.Conclusion rhIL-6 can induce apoptosis of the cell of mouse bladder carcinoma,apoptosis is likely to proceed through the Fas signal transduction pathway.
