食管癌细胞SHEEC中NGAL基因5′端转录调控区的克隆及其顺式作用元件定位分析

【摘要】 目的 :从食管癌细胞SHEEC中克隆NGAL基因 5′端转录调控区并进行顺式作用元件定位分析。方法 :采用PCR法克隆NGAL基因 5′端转录调控区 ,并通过NCBI公共数据库BLAST序列分析鉴定突变。应用KEGG序列数据库对该调控区进行顺式作用元件定位分析。结果 :从SHEEC中获得了NGAL基因 5′端转录调控区 - 112 4~ +6 5区段的克隆 ,该区段序列有 3处点突变 ,在NGAL基因- 112 4~ +6 5区段共鉴定出 78个有效顺式作用元件位点。结论 :多种反式作用因子可能是永生化食管上皮细胞恶性转化中NGAL基因转录增强的重要调控因素

【Abstract】 Objective To clone neutrophil gelatinase associated lipocalin(NGAL)gene 5′ nontranscribed region and to analyse its potential cis acting elements from esophageal carcinoma cell SHEEC.Methods NGAL gene 5′ nontranscribed region was cloned by PCR.After sequencing the base pair mutation character was confirmed by using NCBI/BLAST database.The positions of the cis acting elements from NGAL gene 5′ nontranscribed region were searched by using KEGG/MOTIF database.Results NGAL gene 5′ nontranscribed region (-1 124 to +65) was isolated from SHEEC.This sequence showed base pair exchanges in three cases after alignment with 5′ flanking region of NGAL gene from neutrophil granulocyte(Accession No. X99133).Seventy eight sites of the potential cis acting elements were identified in the fragment(-1 124 to +65).Some of them were the special binding sites of trans acting factor.Conclusions Activation of the several transcription factors in TPA/PKC pathway might be the important regulation factors of increased transcription of NGAL gene in the progress of malignant transformation of human immortalized esophageal epithelial cell.
