聚合酶链反应检测铜绿假单胞菌感染性角膜溃疡的实验研究及临床应用
Experimental study and clinical application of fsat diagnosis of Pseudomonsa aeruginosa corneal ulcer using polymerase chain reaction
【摘要】 目的 :探索聚合酶链反应 ( PCR)技术对铜绿假单胞菌感染性角膜溃疡快速诊断的可行性、优越性及其在临床应用中的价值。方法 :建立 PCR检测标准铜绿假单胞菌方法 ,对兔铜绿假单胞菌感染性角膜溃疡模型研究并应用于临床 ,并与细菌培养做比较。结果 :铜绿假单胞菌扩增出一条长 5 0 4 bp的阳性条带 ,其他常见细菌、人和兔正常角膜组织均为阴性。动物模型 PCR敏感性为 93.8% ,特异性为 10 0 .0 % ;细菌培养敏感性为 6 8.7% ,特异性为 93.7%。临床标本 PCR阳性率为 6 6 .7% ;细菌培养阳性率为 38.1%。结论 :PCR技术对快速检测实验室和临床标本中的铜绿假单胞菌具有重要的应用价值
【Abstract】 Objective:To investigate the feasibility and superiority of fast diagnosis of Pseudomonas aeruginosa corneal ulcer using polymerase chain reaction and its application in clinical diagnosis.Methods:PCR was performed to detect the DNA segment of Pseudomonas aeruginosa.Results:A 504bp product was successfully amplified from Pseudomonas aeruginosa, but not from other bacteria and rabbit or human cells. In model of rabbit Pseudomonas auruginosa corneal ulcer, the sensitivity of PCR was 93.8%, the specificity of PCR was 100%;While the sensitivity of culture was 68.7%, and its specificity of PCR was 93.7%. The sensitivity of PCR and culture were 66.7% and 38.1% in 21 clinical specimens.Conclusion:PCR is of great value to detect Pseudomonas aeruginosa rapidly in experimental and clinical specimen.
