人眼α-晶体蛋白分子伴娘活性的研究
The effects of ageing on human-crystallin’ s molecular Chaperone-like Activity
【摘要】 目的 了解不同年龄人α-晶体蛋白分子伴娘活性,进一步探索α-晶体蛋白的分子伴娘活性与年龄相关性白内障形成间的关系。方法(1)用凝胶过滤法分别分离成年人及老年人水溶性晶体蛋白。在55℃高温条件,观察不同年龄人α-晶体蛋白对β-low晶体蛋白的凝集和变性的抑制程度差异。结果(1)两组不同年龄的人α-晶体蛋白对热诱导的β-low晶体蛋白的凝集和变性的抑制程度有差异,成年人α-晶体蛋白对热诱导的β-low晶体蛋白的凝集和变性的抑制作用比老年人α-晶体蛋白强。结论(1)随着年龄的增加,人α-晶体蛋白的分子伴娘活性呈下降趋势。
【Abstract】 Objective To study with ageing whether α-crystallin lose their molecular chaperone-like ability , through this to better understand the becoming of Age-related cataract.Methods (1) Tow different ages human eye (18-30 years old,60 - 88 years old ) was chosed to do research. The water-soluble crystallin of eyes was purified by Sephadex G - 200 gel filitration. Observe different ability of them to protect β-1ow crystallin from thermal aggregation. Results (1) α-crystallin of 18 - 39 years old eyes has more ability to protect β-low crystallin from aggregation than 60-88 years old. Conclusion (1) With ageing human eye’s α-crystallins gradually lose their molecular chaperone - like ability.
