基于真空渗入遗传转化的菜薹苗态建成方法研究
Study on forming optimum seedling state for vacuum infiltration genetic transformation in B.chinensis var.utilis
【摘要】 采用不同时期播种和人工低温春化处理2种方法,对真空渗入法进行菜薹遗传转化的适宜苗态及其建成方法进行研究.结果表明,菜薹的适宜苗态是:植株已完成花芽分化,花薹未完全抽出,第2,3,4片真叶平均长6.0cm以上,宽3.0cm以上,茎粗4.0mm以上,具8~9片真叶.陕西关中地区适宜抽薹期为4月上旬.2月中旬播种于温室中,4月上旬进行真空渗入最为适宜.不同品种根据冬性强弱,种子低温(4℃)处理12~24d可以形成最适苗态.一叶一心幼苗低温(6℃)处理对苗态有显著影响,是建立适宜苗态的另一有效途径.
【Abstract】 The methods to form optimum plant state were studied for vacuum infiltration genetic transformation in B.chinensiss var.utilis.The results showed that the suitable seedling state is as follows:the flower bud differentiation finished, the internodes starting elongation, the leaf length and width over 6.0 cm and 3.0 cm respectively, stem thickness over 4.0 mm,having eight to nine leaves.Bolting period was best in the first tenday of April planting in Guanzhong area of Shaanxi;It was most suitable to form optimum plant state to sow in greenhouse in the middle tenday of February and to infiltrate in the first and middle tenday of April. The different varieties require 12~24 d low temperature at 4 ℃ for vernalization to form optimum plant state.Low temperature treatment (6℃) seedlings,having significant effect on seedling state,is another effective way for forming optimum seedling state.
