血红蛋白分子氧合过程的构象变化特性
The Features of Conformational Changes of Hemoglobin during the Oxygenation
【摘要】 在分析血红蛋白分子与氧分子结合的相互作用特点的基础上,将氧分子与血红蛋白分子结合的过程看作为协同可逆吸附过程.根据协同吸附过程的特点,利用吸附动力学方法,得到了反映血红蛋白分子氧合过程的氧饱合分数与氧分压之间的关系式.从上述关系式得到了与之自洽的推论,在一定的氧分压条件下,血红蛋白分子处于确定的部分氧合构象态.当氧分压的值从小到大变化时,血红蛋白分子将从脱氧构象态经历一系列中间构象态到达完全氧合构象态.这表明S型的氧合曲线实际显示了单个血红蛋白分子的别构效应.
【Abstract】 On the basis of analyzing the features of interaction between hemoglobin and oxygen molecules, the oxygenation of hemoglobin can be treated as a cooperative adsorption process. According to the properties of coorperative adsorption process,the relations of the fractional saturation with the partial pressure of oxygen can be obtained by means of adsorption kinetics. The conclusion can be rationally drawn from the above formula. It indicates that hemoglobin is at a fixed partial-deoxy conformational state under certain partial pressure of oxygen. As oxygen pressure increases, the conformation of hemoglobin will undergo a series of intermediates from deoxygenated form to oxygenated one. It demonstrates that a typeS curve of oxygenation represents the real allosteric effect that appears in a Hb molecule during oxygenation.
