大豆子叶节组培再生系统与农杆菌介导的基因转化系统的比较研究
THE STUDY OF COMPARING THE TRANSFORMATION SYSTEM OF AGROBACTERIUM-MEDIATED AND REGEERATION SYSTEM OF COTYLEDON NOD OF SOYBEAN CULTURE
【摘要】 不同基因型的大豆子叶节在添加适宜 BA的 MS培养基上培养 ,可从子叶节诱导出高频丛生芽 ,并获得大量再生植株 .通过农杆菌介导法 ,将深黄被孢霉Δ6 - 脂肪酸脱氢酶基因导入到栽培大豆吉林 43和黑农 3 7品种中 .经过在含 5 0 0 mg/L卡那霉素的筛选培养基中连续筛选 ,获得一批转基因植株 .经 PCR检测和 DNA分子杂交分析 ,初步证明外源基因已导入并整合到大豆基因组中 .本文重点阐述了组培再生系统和基因转化系统的内在联系和不同点 .
【Abstract】 High frequency multiple shoot-bud formation from different genotype of soybean cotyledon node inducing culture on a MS-medium supplemented with BA was obtained. Plant can be obtained from multiple shoot-bud. The Δ 6_- fatty acid desaturase gene from Mortitieralla isabellina was transferred into cultivars Jilin 43 and heinong 37 via Agrobacterium infection by inducting with various explant methods. Multiple shoot and regeneration plant were inducted rrom cotyledon node. Kanamycin resistant (kan r) plantlets were obtained by 3-4 cycles screening in selective condition. PCR and Southern blot analysis from the transferred gene plants proved that foreign gene were transferred and integrated into soybean genome.
