大鼠胰腺β肿瘤细胞中胰岛素样生长因子结合蛋白-2和3的表达调控
Expression and Regulation of IGFBP-2 and-3 in Insulin Producing Cell Line of Rats
【摘要】 目的:研究大鼠胰腺β肿瘤细胞(INS-1)中,生长激素(GH)、催乳素(PRL)、葡萄糖和胰岛素对胰岛素样生长因子结合蛋白-2和3(IGFBP-2、3)的表达调控,探索IGFBP-2和3对维持正常β细胞生理功能的作用。方法:RNA印迹法检测INS-1细胞中IGFI3P-2和3的mRNA表达情况及蛋白免疫印迹法分析INS-1细胞中IGFBP-3和2蛋白表达。结果:在INS-1细胞中,GH和PRL调控IGFBP-3 mRNA和蛋白表达。葡萄糖对IGFBP-2 mRNA调节作用不同:高浓度(10 mmol/L)葡萄糖条件下,IGFBP-2 mRNA表达被抑制;低浓度(1.0~5.6 mmol/L)葡萄糖条件下,IGFBP-2 mRNA有表达。葡萄糖浓度为5.6 mmol/L时,加入GH孵育16 h,IGFBP-2 mRNA表达被抑制。外源胰岛素抑制 IGFBP-2 mRNA的表达。结论:GH和PRL可以调控IGFBP-3的mRNA和蛋白表达,IGFBP-2 mRNA表达主要通过胰岛素调控。
【Abstract】 Objective:To study the effects of GH, PRL, glucose and insulin on IGFBP-3 mRNA and IGFBP-2 mRNA expression in INS-1 cell. Methods: The expression of tGFBP-2,3 mRNA was detected by Northern blot and IGFBP-3 proteins was detected by method of Western blotting. Results: The expression of IGFBP-3 mRNA was up-regulated by bovine GH(bGH), rat PRL(rPRL) in the INS-1 cells. The glucose had variant effect on IGFBP-2 gene expression. On high concentration of glucose (10 mmol/L), the expression of IGFBP-2 mRNA was suppressed. On low concentration of glucose (1.0-5.6 mmol/L), expression of IGFBP-2 mRNA was normal. After incubation 16 h with exogenous GH the expression of IGFBP-2 was inhibited. The expression of IGFBP-2 mRNA was inhibited by exogenous insulin at low glucose concentration. Conclusion:The expression of IGFBP-3 mRNA is regulated by GH and PRL and the expression of IGFBP-2 mRNA is mainly regulated by insulin.
