兔视网膜色素上皮细胞培养方法的改进

【摘要】 目的:寻求一种更好的培养兔视网膜色素上皮(RPE)细胞的方法。方法:在无菌滤纸上剪开眼球,固定眼杯,用胰酶直接消化视网膜色素上皮面,收取RPE细胞。将初次换液时间由取材后48 h延长至72 h,记录改变换液时间对细胞数量及细胞形态等的影响。用多巴(DOPA)染色和免疫组织化学对细胞鉴定。结果:采用改进后方法收取的细胞呈分散均匀生长。延长初次换液时间为72 h后获得的细胞增多,原代细胞长至融合状态需要的时间缩短,色素颗粒脱失现象减轻。结论:采用该方法更适于RPE细胞的收获和培养。

【Abstract】 Objective: To improve the cultivating method of retinal pigment epithelium cells (RPE) in rabbits. Methods:The eye ball was bisected along the equator on a sterile filter paper. The anterior portion of eye was discarded, then the posterior eye cup was fixed in a rubber bysma. The single cell was collected directly by trypsinization. The first time for the medium to be changed was prolonged from 48 h to 72 h. The RPE cells were identified by DOPA staining and immunohistochemistry. Results: RPE cells grew in well-distributed way rather than in colony in this experimental condition. Prolonging first changing medium time was found to increase the number of cells. RPE cells reached confluence more quickly and the melanin disappeared more slowly. Conclusion: The modified method is more suitable for cultivating RPE cells in rabbits.
