As2O3诱导宫颈癌HeLa细胞凋亡与细胞内端粒酶活性关系的研究

【摘要】 目的 :探讨As2 O3 对HeLa细胞的诱导凋亡作用及对HeLa细胞端粒酶活性的影响。方法 :采用不同浓度的As2 O3 作用于人宫颈癌HeLa细胞不同生长时间段 (2 4、48、72h)后 ,显微镜下观察细胞凋亡 ,四甲基偶氮唑盐比色法 (MTT法 )测定细胞生长抑制情况 ,TRAP ELISA方法测定细胞内端粒酶活性变化。结果 :2 μmol·L-1的As2 O3 处理HeLa细胞 48h后可降低细胞存活率 ,诱导细胞凋亡 ,细胞内端粒酶活性比对照组明显下降。随着As2 O3 浓度增加、作用时间延长 ,作用效果越明显。结论 :As2 O3 诱导HeLa细胞凋亡可能与其抑制细胞内端粒酶活性有关。

【Abstract】 Objective To study the inductive function of As 2O 3 on apoptosis of HeLa cells and its influence on telomerase activity.Methods HeLa cells were treated with As 2O 3 of various concentrations and observed at 24,48 and 72?h for morpho?logical alterations.The effect of As 2O 3 on HeLa cells was determined by MTT assay,and telomerase activity in HeLa cells was determined by TRAP?ELISA.Results 2?μmol·L -1As 2O 3 induced apoptosis of HeLa cells at 48?h and reduced their survival,and could also suppress telomerase activity in the cells under proper controls.It was found that the higher the As 2O 3 concentration was and the longer the treating time was,the better the effect was.Conclusion As 2O 3 inhibiting the telomerase activity in HeLa cells may be involved in the mechanism of the cell apoptosis.
