茶多酚对高糖所致人肾小球系膜细胞活性氧产生的影响
Effect of tea polyphenols on high glucose-induced reactive oxygen species in human mesangial cells
【摘要】 目的 :探讨高糖对人肾小球系膜细胞活性氧产生的影响以及茶多酚对其的干预作用。方法 :应用比色法检测高糖作用后培养系膜细胞上清液中超氧化物歧化酶 (SOD)、谷胱甘肽过氧化物酶 (GSH PX)的活力和丙二醛 (MDA)、超氧阴离子自由基 (O2-· )的水平 ,以及加入茶多酚后各指标的改变。结果 :高糖可降低系膜细胞培养上清液中SOD、GSH PX的活力 ,增加MDA和O2 -· 的含量 ;茶多酚可提高上清液中SOD和GSH PX的活力 ,降低MDA和O2 -· 的含量。结论 :高糖可导致人肾小球系膜细胞活性氧产生增多 ,而茶多酚可进行有效干预
【Abstract】 Objective To investigate the effect of high glucose on reactive oxygen species (ROS) in human mesangial cells and the intervention of tea polyphenols(TPs).Method Colorimetric assay was used to detect SOD、GSH?PX、MDA、O 2 -· in the supernatant of high glucose culture of human mesangial cells treated with or without TPs.Results SOD and GSH?PX were lowered,while MDA and O 2 -· increased in the supernatant of high glucose culture without TPs.However,SOD 〔( 32.507± 3.368)NU·ml -1 〕 increased with TPs compared with that only with high glucose,GSH?PX〔(56.233±2.808) NU· ml -1 〕increased as well,while MDA〔(3.280±0.149)μmol·L -1 〕and O -· 2〔(29.751±2.528)N·L -1 〕decreased.Conclusion High glucose contributes to an increase in ROS produced by human mesangial cells and TPs can better control high glucose induced oxidative stress.
