小菜蛾对杀虫双抗性遗传的RDA分析

【摘要】 采用代表性差异分析法 (RDARepresentationaldifferenceanalysis)初步研究了小菜蛾两个品系 ,敏感品系和抗杀虫双近等基因系之间基因组的差异 .用敏感品系作为驱动扩增子 (driver) ,抗杀虫双近等基因系作为检验扩增子 (tester) ,通过四轮消减杂交后 ,得到两个差异片段 ,范围在 1 50bp至 30 0bp之间 ,初步判断这些片段可能与小菜蛾杀虫双抗性的遗传分子基础相关

【Abstract】 The genomic difference between plutalla xylostella susceptible strain and dimehyo resistance near isogenic strain was preliminarily analyzed by RDA (Representational Difference Analysis) technique. In RDA system, the driver amplicon is susceptible strain while the tester amplicon is dimehyo resistance near isogenic strain. After four rounds of subtractive hybridization, two different products were obtained and their sizes were about 150?bp~300?bp. Preliminary judgement revealed that these segments might be associated with genetic molecular mechanism of dimehyo resistance in diamondback moth.
