受白叶枯病菌诱导的水稻抗病相关基因rgi97的部分序列克隆

【摘要】 利用mRNA差异显示技术从水稻中克隆到一个受白叶枯病菌诱导的基因片段 (rgi 97)。根据RT PCR结果 ,rgi97具有诱导表达特性 ,在水稻受白叶枯病菌侵染后 2 4h时检测不到 ,36h时有微弱表达 ,而到 4 8h时表达增强。通过3′RACE技术获得了该片段 3′端约 970bp的序列。相似性搜索表明 ,包含 3′末端的rgi 97可能的编码产物与拟南芥一个未知蛋白有一定相似性 ,达 92 % ,提示该基因可能与水稻对白叶枯病的抗性有关。

【Abstract】 Cloning and functional analysis of rice genes involved in pathogen resistance and induced by Xanthomonas oryzae pv.oryzae(Xoo) is an important step towards understanding molecular mechanisms of rice resistance to Xoo.The 465 bp partial sequence of a gene,named rgi 97,was obtained from rice by differential display of mRNA isolated from rice plants inoculated with Xoo.The rgi 97 was expressed in plants infected by bacteria but not in plants uninfected.Transcript of rgi 97 was not detected in plants of rice varieties IR24 and IRBB4 until 36 h after inoculation,and reached maximal amount by 48 h.The 3′-terminal sequence of rgi 97 was cloned by 3′-rapid amplification of cDNA end(RACE),and sequenced.Blast search indicated that the deduced amino acids of rgi 97 shared homology with an unknown protein from Arabidopsis thaliana.These results suggested that rgi 97 may be a novel gene involved in rice resistance to Xoo.Further study is required to determine nature and function of the gene.
