家蚕蛹表达的乙型肝炎表面抗原中蛋白口服免疫原性的研究
Study on oral immunogenicity of HBsAg (PreS2+S)expressed in silkworm pupa
【摘要】 目的 探讨家蚕蛹表达的重组乙型肝炎表面抗原中蛋白 [rHBsAg(PreS2 +S) ]口服免疫原性。方法 选择 4周龄的SD大鼠 ,将重组病毒感染后的家蚕蛹 ,制成口服剂型灌胃大鼠 ,ELISA方法检测抗 HBs抗体及抗 PreS2抗体。结果 在灌胃重组蛋白组中有 4 0 %鼠体血清内检测到了特异性抗 HBs抗体及抗 PreS2抗体 ,抗体应答反应持续 2~ 3周 ;口服免疫后抗体转阳的SD鼠加强免疫后能产生较强的二次免疫应答。进一步研究表明 ,口服家蚕蛹表达的rHBsAg(PreS2 +S)具有加强免疫的效果。对酵母重组疫苗单剂腹腔注射过的SD鼠 ,用家蚕蛹表达的rHBsAg(PreS2 +S)灌胃加强免疫后 ,能诱导鼠体迅速产生较强的抗 HBs抗体。结论 家蚕 BmNPV表达载体系统将有望开发成为新型的口服乙型肝炎疫苗表达系统
【Abstract】 Objective To determine the oral immunogenicity of silkworm pupa-derived rHBsAg (PreS2+S).Methods Gavaging with silkworm pupae producing rHBsAg (PreS2+S) to rats at 4 weeks of age and detecting anti-HBs and anti-PreS2 antibodies with ELISA. Results Anti-HBs and anti-PreS2 antibodies were detected in 40 percent rats gavaged. A stronger secondary antibody response was obtained only in the rats of antibody conversion after the booster injection. We also found that rHBsAg(Pre+S)-silkworm pupae gavaged to rats could be used to boost an immuneresponse. Rats primed with a single yeast-derived rHBsAg and gavaged silkworm pupae containing rHBsAg(Pre+S) developed immediate antibody responses.Conclusion Silkworm-BmNPV expression vector system will widen the scope, as novel expression system to develop edible HB vaccine.
