TH1类细胞因子对结核病患者产生单核细胞趋化蛋白1的调节作用
Effect of TH1 cytokines on the production of MCP-1 by peripheral blood mo no nuclear cells from patients with tuberculosis
【摘要】 目的 探讨TH1 类细胞因子干扰素γ(IFN γ)、白细胞介素 2 (IL 2 )对结核病患者外周血单个核细胞 (PBMC)分泌MCP 1的影响。方法 将 1 2例结核患者和 8例健康人PBMCs在LPS诱导前 ,用IL 2、IFN γ进行预刺激 1h ,采用ELISA法检测各组培养上清液中MCP 1的水平。结果 与单纯用LPS组相比 ,加入IL 2、IFN γ能明显上调结核患者及健康人PBMC的MCP 1分泌 ,且呈现浓度依赖性 ,两者联用具有协同效应。结论 TH1 类细胞因子IL 2、IFN γ通过上调MCP 1 ,使单核细胞、淋巴细胞向炎症部位趋化 ,参与组织炎症及肉芽肿形成 ,从而发挥保护性的免疫效应
【Abstract】 Objective To investigate the effect of T H1 cytokine s on t he production of MCP 1 by peripheral blood mononuclear cells(PBMCs) from patien ts with tuberculosis. Methods PBMCs were isolated from twelve t uberculosis patients and eight healthy individuals and then cultured. The cultur es were pretreated with IL 2,IFN γ for 1 hour before being stimulated with L PS. The levels of MCP 1 were detected with ELISA. Results Comp ared with the LPS group, IL 2 and IFN γ significantly up regulated the prod uction of MCP 1 in the patients and the healthy individuals and the effects wer e dose dependent. The effects of IL 2 and IFN γ on LPS induced MCP 1 secre tion were simply addi tive. Conclusions The T H1 dri ved c ytokines IL 2 and IFN γ up regulated the production of MCP 1,which recruite d the mon ocytes and lymphocytes to the site of infection. Thus it plays a key role in the tissue inflammation and granuloma formation.
