CIK细胞对胃癌OCUM-2MD3细胞体内外杀瘤活性的实验研究

【摘要】 目的 研究正常人细胞因子激活的杀伤细胞 (Cytokine inducedkiller,CIK)对人胃癌细胞株OCUM 2MD3的体外细胞毒活性以及对胃癌腹膜移植模型的体内抗肿瘤作用。方法 取正常人外周血单个核细胞 (Peripheralbloodmononuclearcell,PBMC) ,加入γ IFN、IL 2、抗 CD3单抗和IL 1,体外诱导成CIK细胞 ,用流式细胞仪对细胞作动态表型分析 ,用MTT法测定CIK细胞体外对OCUM 2MD3的细胞毒活性 ,并与CD3AK作对比。在Balb/c裸小鼠腹腔内接种OCUM 2MD3细胞 ,观察CIK细胞对荷瘤鼠的抑瘤作用。结果 CIK细胞在培养 2周左右获得大量增殖 ,CD3+ CD5 6 + 双阳性细胞大量增殖达 10 0 0倍以上 ;体外实验证明 ,CIK细胞对OCUM 2MD3有明显的细胞毒活性 ,对比CD3AK其抑瘤率为 85 %与 6 2 .8% (P <0 .0 5 )。体内实验表明 ,CIK细胞能够显著抑制癌性腹水的生长 ,其抑瘤率可达 10 0 % ;肿瘤标志物CEA含量明显降低。结论 CIK细胞是一种新型、高效的免疫活性细胞 ,具有较强的体内外抗胃癌细胞活性 ,具有明显的抑制癌性腹水的生长的作用 ,有可能用于临床上晚期胃癌的过继性免疫治疗

【Abstract】 Objective To investigate th e cytotoxicity of CIK (Cytokine induced killer) cells from healthy donor in v itro and their anti tumor effects in Balb/c nude mice bearing OCUM 2MD3 cell s strain of ascites tumor from gastric carcinoma.Methods Periph eral blood mononuclear cells(PBMC) from healthy donors were induced to be come CIK c ells by cytokine in vitro incubation. The CIK cells were analyzed on differe nt t ime points by FACS; Compared with CD3AK cells, the cytotoxicity in vitro by MTT assays and the antitumor activity of the CIK were evaluated in Balb/c nude mice bearing OCUM 2MD3 cells from carcinoma of stomach.Results Flow cytometry analysis showed that CD3 +CD56 + T cells expanded 1 000 fold after 2 weeks in CIK culture; CIK cells possessed a higher antitumor cytotoxic activit y on OCUM 2MD3 cells in vitro than CD3AK cells; In addition, CIK cells had a stronger suppressive effect on tumor growth in Balb/c nude mice bearing OCUM 2MD3 tumor compared with CD3AK cells in vivo (Median inhibitory rates 85% vs 62.8%, P <0.05).Conclusion CIK cells are highly e fficient cytolytic effector cells which have a stronger significant suppression against growth of carcinoma of stomach in vitro and in vivo . The result provides an experimental basis for CIK to clinical application as a adoptive imm unotherapy. [
