DNA末端原位标记技术研究慢性再生障碍性贫血骨髓CD34+细胞凋亡
Study on Apoptosis of Bone Marrow CD34+ Cells in Chronic Aplastic Anemia Patients by DNA in Situ End Labelling
【摘要】 目的 观察慢性再生障碍性贫血 (CAA)患者骨髓CD3 4+ 细胞凋亡状况 ,探讨CAA可能的发病机制。方法 采用单克隆抗体 -免疫磁珠分离系统 (MACS)分离纯化 39例CAA患者的骨髓CD3 4+ 细胞 ,用DNA末端原位标 (TUNEL)技术分析骨髓CD3 4+ 细胞原位凋亡状况。结果 CAA患者骨髓细胞的凋亡率为 (39.2 4± 12 .70 ) % ,健康对照组为 (8.5 0± 2 .30 ) % ,组间比较差异有显著性 (P <0 .0 1)。凋亡细胞可见核染色质皱缩、凝聚 ,但未见到核碎裂现象及典型的凋亡小体。结论 CAA患者骨髓CD3 4+ 细胞的过度凋亡是其质或 /和量缺陷的原因之一
【Abstract】 Objective To observe apoptosis of bone marrow CD 34 + cells in chronic aplastic anemia (CAA) patients and to investigate the pathogenesis of CAA.Methods Bone marrow CD 34 + cells were separated from 39 CAA patients with immunomagnetic beads(MACS)and analysed with TUNEL labelling.Results Apoptosis rate of CD 34 + bone marrow cells in CAA patients was (39.24.±12.70)%,while in the control group (8.50±2.30) so that there was a significant difference between two groups (P<0.01).Chromatin condensation was observed in apoptosis cells while neither nuclear crashing nor apoptosis body was found.Conclusion Over apoptosis in CD 34 + bone marrow cells of CAA is one of the reasons why the quality or number of CD 34 + cells declines
