晶状体上皮细胞培养特征及细胞因子对其增殖影响的实验研究

【摘要】 目的 探讨体外培养的牛晶状体上皮细胞的培养方法及特性 ,并研究γ -干扰素 (γ -inter feron ,γ -IFN)对晶状体上皮细胞增殖的抑制作用。方法 牛晶状体上皮细胞原代与传代培养。第 2代培养细胞中添加γ -IFN ,浓度 1 0 1 - 1 0 5U /ml,通过甲基噻唑基四唑 (MTT)测定法 ,观察γ -IFN对晶状体上皮细胞的影响。结果 γ -IFN在体外有抑制牛晶状体上皮细胞增殖作用 ,并以 1 0 4 U /ml最为明显。结论 γ -IFN可抑制晶状体上皮细胞增殖作用 ,因而能抑制后发障的形成

【Abstract】 Objective To elucidate the method of culturing Bovine lens epithelial cells in vitro and the characteristics of the cells and to investigate the inhibition of γ-interferon(γ-IFN) on the proliferation of bovine lens epithelial(BLECs).Methods Primary culture and subculture of BLECs were established in vitro. γ-IFN at concentrations from 10 1U/ml-10 5U/ml were added to the epithelial cells of second passage. The inhibitive effects of γ-IFN on proliferation of BLECs were measured with methyl thiazolyl tetrazolium(MTT) assay method.Results After treatment withγ-IFN, lens epithelial cells showed marked inhibition depending on the concentration of γ-IFN from 10 3U/ml to 10 5U/ml,especially in 10 4U/ml of γ-IFN.Conclusion γ-IFN plays a significant role in inhibiting the proliferation of BLECs.
