滩羊生长激素基因PCR-RFLPs反应体系的优化
OPTIMIZATION OF PCR-RFLPS REACTION SYSTEM FOR TAN SHEEP GH GENE ANALYSIS
【摘要】 以宁夏滩羊基因组DNA为模板 ,研究影响滩羊PCR -RFLPs扩增的因素 ,实验结果表明 :特异性引物的浓度、Mg2 + 浓度、模板DNA的完整性 ,浓度、纯度、TaqDNA聚合酶、变性温度、退火温度和循环次数等诸多因素对PCR -RFLPs扩增影响较大。为此我们对PCR -RFLPs的反应体系和反应程序进行了研究和浓度梯度对比试验 ,建立了滩羊GH基因PCR -RFLPs扩增的反应体系和操作技术
【Abstract】 The study on factors affecting amplification of PCR-RFLPs for Tan sheep of Ningxia carried out by using Tan sheep genetic DNA as a template The result showed that the concentrations of specific primer Mg ++ , Taq polymerase and the purity and completion of template DNA, denaturing temperature, annealing temperature, the number of cycles played an important role in PCR-RFLPs analysis We studied and tested the concentration step’s contrast for PCR-RFLPs reaction system and reaction process A technical system for Tan Sheep GH gene PCR-RFLPs was estab-lished
