【摘要】 目的　探讨淋巴瘤中存在肿瘤干细胞的可能性。方法　收集 2例新鲜反应性增生淋巴结和 1例外周T细胞淋巴瘤标本进行原代培养 ,对培养 1d、12d、19d的细胞进行S P法免疫组化染色。同时用Raji细胞系接种 3只SCID鼠 ,在光镜、电镜下观察成瘤组织结构变化。并观察Raji细胞系、成瘤组织及再培养 1周成瘤组织细胞免疫表型的变化。 结果　淋巴结细胞原代培养 12d、19d后剩余极少量细胞表达CD8、CD3、CD2 0、TDT ;在细胞系及接种成瘤组织中 ,免疫组化查见少量CD5、TDT阳性细胞 ;Raji细胞系接种前后电镜观察见不同比例中心细胞及中心母细胞。 结论　反应性增生淋巴结中存在淋巴结干细胞 (或者长期记忆细胞 )。细胞系及接种成瘤组织中CD5、TDT阳性细胞可能是肿瘤干细胞 ,进一步提示肿瘤干细胞存在的可能性更多还原

【Abstract】 Purpose To explore the possibility that there were cancer stem cells in lymphoma. Methods The fresh tissues of 2 reactive lymph node cases and one lymphoma were collected for primary culture. The culture cells were investigated by immunohistochemistry in 1,12, and 19 days. Raji cell lines which were injected into 3 SCID mice grew up into neoplasms, and the neoplasms were observed by light and electron microscope. Raji cells, the tumor tissues and the tumor cells after culturing for a week were observed by immunohistochemistry. Results Cells expressing of CD3, CD8, CD20 and TDT were found in lymph nodes, which were cultured for 12, 19 days. There were a few CD5, TDT positive cells in Raji cell lines and the injected tumor tissues. The centrocytes and centroblasts with variable number were seen in Raji cell lines and the injected tumors by electron microscope. Conclusions There are cancer stem cells (or long memory cells)in reactive lymph node. It is possible that CD5, and TDT positive cells are cancer stem cells in Raji cell line and injected tumors, further indicating the possibility of cancer stem cells existed in tumor.更多还原
