酸性品红与蛋白质结合反应的研究及应用
STUDY ON THE COMBINATION OF PROTEINS WITH ACID FUCHSIN AND ITS ANALYTICAL APPLICATION
【摘要】 在 pH 1.4的Clark Lubs缓冲介质中 ,蛋白质与酸性品红反应形成结合物 ,该结合物的最大吸收波长为 5 76nm ,与酸性品红相比 ,红移了 34nm。据此拟定了分析方法 ,制定了牛血清白蛋白 (BSA)、人血清白蛋白 (HAS)、γ 球蛋白 (γ G)和酪蛋白的工作曲线 ,测定的摩尔吸光系数均在10 5数量级 ,用于人血清中总蛋白的测定 ,结果与溴甲酚绿 (BCG)法基本一致。
【Abstract】 In a Clark Lubs buffer medium of pH 1.4, protein combines with acid fuchsin to give a product having its absorption maxima at 576nm which is red shifted 34nm from the absorption maxima of acid fuchsin. Standard curves of 4 kinds of proteins, i.e. the bovine serum albumin (BSA), the human serum albumin (HSA), the γ globulin (γ G) and the casein, were prepared under the optimum reaction condition. The molar absorptivities were found to be 3.66×10 5L·mol -1 ·cm -1 , 3.83×10 5L·mol -1 ·cm -1 , 1.72×10 5L·mol -1 ·cm -1 and 5.41×10 5L·mol -1 ·cm -1 respectively. This method has been applied to the determination of total proteins in human serum giving results in comformity with those obtained by the bromocresol green (BCG) method.
