【摘要】 用沸水提取白阿魏菇菌丝体多糖 ( PNMP) ,经 DEAE-cellulose 3 2及 Sephadex G-1 0 0柱层析得纯化多糖 PNMP,用水杨酸法测 PNMP对· OH的清除作用 ;用硫代巴比妥酸法测小鼠肝组织丙二醛 ;用分光光度法测小鼠红细胞溶血和肝线粒体肿胀 ;测 PNMP对连苯三酚自氧化体系的抑制作用 .结果表明 PNMP能明显地清除· OH,抑制· OH所致的丙二醛增长 ,减少· OH所致红细胞溶血和线粒体的肿胀 ,并抑制连苯三酚自氧化 ,所以 PNMP有抗氧化活性更多还原

【Abstract】 A Pleurotus nebrodensis mycelium polysaccharide (PNMP) was extracted with boiling water from Pleurotus nebrodensis mycelium.The polysaccharide was separated by using DEAE-cellulose and purified by using Sephadex G-100 column chromatography.The scavenging effect of PNMP on hydroxyl radical (·OH) was studied by the salicylic acid method.To study the anti-oxidative effect of PNMP on ·OH,the content of MDA in mice liver was measured by TBA assay.The hemolysis extent of red blood cells and the swelling extent of mitochondria of liver were detected by spectrophotometric methods and self-oxidation inhibition was studied using the 1,2,3 phentriol.The result showed that PNMP had the ·OH scavenging activity.It could inhibit the generation of MDA and self-oxidation,so as to decrease the swelling of mitochondria and the hemolysis of red blood cells induced by ·OH .The conclusion was that PNMP had the effect of antioxidation.更多还原
