HPLC法测定血浆中托瑞米芬浓度

【摘要】 目的:建立简便可靠的托瑞米芬(Tor)体内浓度的测定方法。方法:以吲哚美辛为内标,蛋白沉淀法进行预处理,用HPLC紫外检测,C18(150mm×4.6mm,5μm,流动相:乙腈-0.1mol·L-1乙酸铵(65:35,V/V),流速1.0mL·min-1,检测波长277nm。结果:Tor保留时间为9.1min,最低定量限为0.5μg·mL-1,线性范围0.5~10 μg·mL-1,r=0.9997,处理后血浆样品稳定性良好,平均批内、批间RSD均<4%。结论:该方法符合相应要求,适用于基础医学研究和Tor人体血药浓度检测。

【Abstract】 AIM: To develop a simple and feasible method for the measurement of toremifene in human plasma. METHODS: Protein precipitation and HPLC-UV detection were used. The analysis column was C18(150 mm×4.6 mm, 5 μm). The mobile phase consisted of acetonitrile-ammonium acetate (0.1 mol·L-1) (65/35, V/V) with the flow-rate of 1.0 mL·min-1. Indomethacin was used as internal standard and the sample was detected at 277 run. RESULTS: The peak of toremifene was spiked at 9.1 min and the limit of quantitation was 0.5μg·mL-1. The concentrations range of standard curve was from 0.5 μg·mL-1 to 10.0μg· mL-1. The plasma sample’s stability was good after treatment and average within-run precision and between-run presicion were less than 4% . CONCLUSION: This methods is suitable for basic medical research and can be used to clinical drug monitoring of toremifene.
