【摘要】 目的:观察大鼠局灶性脑缺血皮质白细胞介素1β(IL-1β)mRNA及其受体和受体拮抗剂表达的动态变化。方法:采用尼龙线栓法大鼠脑缺血模型,逆转录PCR方法观察缺血后其相对表达量。结果:缺血侧IL-1βmRNA在各缺血时间点皆有表达,12~24 h达高峰;IL-1RA在缺血48 h表达最强;IL-RI在缺血侧和缺血对侧均稳定表达(包括假手术组);IL-RⅡ仅在缺血侧有表达,24~48 h表达较高。结论:局灶性脑缺血时IL-1βmRNA表达增高,其受体及受体拮抗剂mRNA表达亦增高;表达程度与时相的差别说明它们在脑缺血中可能起着不同的作用。

【Abstract】 Aim:To investigate the gene expression of IL-1β and its receptor (IL-1Rs) and receptor antagonist (IL-1RA) after focal stroke in rats.Methods: Quantitative reverse transcription and polymerase chain reaction (RT-PCR) technique was used to examine the mRNA expression profile of IL-1β, EL-1RA and two EL-1R isoforms after permanent occlusion of the middle cerebral artery(MCAO) in rats. Results: IL-1β mRNA expressed very early in ischemic cortex at all time points and greatly increased between 12 hours and 24 hours; the expression of EL-1RA mRNA peaked at 48 hours;IL-RI mRNA was relatively high expressed in normal cortex and was further elevated late after ischemic injruy. In contrast, IL-RⅡ mRNA only expressed in ischemic cortex, reaching peak levels between 24 hours and 48 hours. Conclusion:The gene expression of IL-1β and its receptor (IL-1Rs) and receptor antagonist(IL-1RA) after focal stroke were all elevated. Differential expression of IL-1β, IL-1RA, IL-R I and IL-RⅡ mRNAs after focal stroke may suggest a distinct role(s) for each component of the IL-1 systems in ischemic injury.
