胶原/纳米磷酸三钙复合人工骨修复骨缺损区TGF-β的表达

【摘要】 目的 :观察胶原 (Collagen ,简称Co) /纳米磷酸三钙 (Nano -TricalciumPhosphate ,简称N -TCP)复合人工骨植入骨缺损区转化生长因子 -β(TGF -β)的分布、刺激引导成骨细胞聚集、促进骨创愈合的能力。 方法 :家兔 2 4只 ,颅骨制成直径 8.0mm圆形洞穿缺损 ,左侧植入Co/N -TCP复合人工骨作为实验组 ,右侧植入非纳米Co/TCP人工骨作为对照组 ;于 2、4、8、12周后处死各组动物取材 ,TGF -β免疫组织化学染色。 结果 :TGF -β免疫组织化学染色显示 :成骨细胞、新生骨细胞大量TGF -β阳性表达 ,细胞间质及纤维组织中散在的TGF -β阳性反应 ;近骨床区各期内两组间TGF -β阳性表达程度无明显差异 ,远骨床区Co/N -TCP组TGF -β阳性表达高于Co/TCP组。结论 :Co/N -TCP复合人工骨在修复骨缺损区可有效刺激引导TGF -β表达 ,促进骨缺损的早期修复。

【Abstract】 Objective:To observe the distribution and function of transforming growth factor-beta (TGF-β) when nano-composite artificial bone (Co/N-TCP) were implanted the region of bone defect. To study the ability of Co/N-TCP recruiting osteoblasts to precipitate the repair of bone defect.Method:Twenty-four domestic rabbits were operated on bilateral cranial bone to create diameter 8.0mm experimental bone defect through the whole bone. On the left, Co/N-TCP was respectively implanted as treatment group. On the right, Co/TCP was also implanted as control group.At 2 W,4 W,8 W,12 W after operation, all animals were sacrificed and implant materials with surrounding bone were taken out. The specimens were collected and treated by histological method. Then immunohistochemical staining of TGF-βin the specimens was attained by avidin-biotin complex method (SABC) to observe the distribution of TGF-β. Result:The inmmunohistochemical observation displayed that a great deal of TGF-βstained existed in osteoblasts and immature osteocytes . Diffused TGF-β positive staining particle appeared in the mesenchymal and fibrous-tissue. There was no significant distinction about the TGF-βpositive staining between two groups in the medial region to original osseous beds in different time . However,in the defected region distal original osseous bed, the TGF-β positive degree in the Co/N-TCP group was significantly higher than that of the control group . Conclusion:The Co/N-TCP has good bioactivities and ability of stimulating and conducting TGF-βto aggregate and precipitate the healing of bone defect.It is a hopeful repairing material for oral-maxillofacial implantation.
