肺癌组织中LRP、GST-π和p53的表达及相关性研究
Study of the Expression of Lung Resistance Protein Gene Glutathions-S-transfetrase-π and P53 Protein in Lung Carcinoma
【摘要】 目的 探讨肺耐药蛋白 (L RP)基因、胎盘型谷胱甘肽 S转移酶 (GST- π)及 p5 3蛋白在肺癌组织中的表达及相关性。方法 应用免疫组化 S- P法检测 5 8例术前未经化疗的肺癌组织中 L RP、GST- π及 p5 3蛋白的表达情况。结果 L RP、GST-π及 p5 3在 5 8例肺癌组织中的阳性表达率分别为 5 5 .17%、6 0 .34%和 74 .14 % ,L RP的表达在不同的组织类型中以腺癌最高 ,达 78.5 7% ,它与鳞癌组相比差异有显著性 (p<0 .0 5 ) ;男女组相比 ,女性组表达率达 78.95 ,差异亦有显著性 (p<0 .0 5 ) ,而与肿瘤的大小及淋巴结转移均无关 (p>0 .0 5 ) ;GST- π的表达在淋巴结转移阳性组高达 78.5 7% ,与阴性组比较差异有显著性 (p<0 .0 5 ) ,而与肿瘤的类型、大小及男女性别无关 (p>0 .0 5 )。 p5 3蛋白在淋巴结转移阳性组为 89.2 9% ,与阴性组相比差异有显著性 (p<0 .0 5 ) ,而 p5 3的表达与肿瘤的类型、大小及性别无明显关系 (p>0 .0 5 ) ;将 L RP、GST-π及 p5 3蛋白的表达进行相关性比较发现 GST-π及 p5 3蛋白的表达关系密切 (p<0 .0 5 )。结论 L RP的表达与肺癌 ,尤其是肺腺癌的耐药有关 ,GST-π及 p5 3蛋白的高表达与肺癌的淋巴结转移相关 ,两者的表达关系密切 ,并可作为肺癌病人预后判断的一个指标
【Abstract】 Objective To explore the expression of lung resistance protein (LRP), glutathione S transferas π (GST π), p53 protein and their relevance in lung carcinoma.Methods Fifty eight patients with lung carcinoma, who had not been treated by chemotherapy before surgery, were studied for the expression of LRP?GST π and p53 protien using immunohistochemical S P methed.Results The frequency of LRP?GST π and p53 was 55.17%, 60.34%, and 74.14%, respectively. The positive expression rate of LRP (78.57%) was significantly higher in adenocacinoma than squamous cell carcinoma (p<0.05) and the positive rate of LRP were higher in woman (78.95%) than man (p<0.05). The positive rate of GST π and p53 in lung carcinoma with lymph node metastasis (70% and 89.29% respectively) were significantly higher than that without lymph node metastasis (p<0.05 ). There was a close relationship between the wxpression of GST π and p53 in lung carcinoma (p<0.05).Conclusion The expression of LRP may be related to drug resistance of lung carainoma. The strong expression of GST π and p53 protein is related to lymph node metastsis of lung carcinoma, which is a better predicitive factor for prognosis in lung cacinoma .
