0℃对CPD-A液保存全血脂质过氧化反应的影响
Effects of different temperatures on lipid peroxidation in human blood preservation with CPD-A
【摘要】 目的 :探讨降低保存温度对血液保存过程中脂质过氧化反应和红细胞变形性 (ED)的影响。方法 :采用化学比色法分别于保存 1d、7d、2 1d、35 d检测 CPD- A液 0℃保存全血红细胞及血浆丙二醛 (MDA )含量 ;红细胞及血浆超氧化物歧化酶(SOD)、全血过氧化氢酶 (CAT)、全血谷胱苷肽过氧化物酶 (GPD)活性 ,血浆总抗氧化能力 (AOA) ,全血谷胱苷肽 (GSH)含量。结果 :2 1d起 0℃保存全血的红细胞及血浆 MDA含量低于 4℃保存 ;2 1d起 0℃保存红细胞 SOD及血浆 SOD活力 ,血浆CAT活力及 AOA、全血 GPD活性及 GSH含量高于 4℃保存 ;2 1d起红细胞滤过指数 (IF)低于 4℃保存 (P <0 .0 5 )。结论 :红细胞的保存损伤与血液的过氧化反应有关 ,降低保存温度可在一定程度上减轻血液的过氧化反应 ,提高红细胞变形能力
【Abstract】 Objective and Methods: In order to study the effect of decreasing the preserving temperature on the quality of blood,the oxidative and antioxidative metoblism and erythrocyte deformablity (ED) were detected by measuring the content of malondialdehyde(MDA) in erythrocyte membrane and plasma, the superoxide dismutase(SOD) activity of erythrocyte and plasma, glutathione peroxidase (GSH-Px), cataiase(CAT) activity of blood,the antioxidite ability(AOA)of plasma and the erythrocyte filtration(IF) at 0℃ and 4℃ during blood storage. Results:Following the prolonged preserving time, the content of MDA in erythrocyte and plasma greatly increased compared with fresh blood(P<0.05,P<0.01),the SOD activities of erythrocyte and plasma ,GSH-Px and CAT activities of blood, AOA of plasma were decreased significantly than that of fresh blood (P<0.05)and the erythrocyte IF was greatly increased compared with fresh blood(P<0.05) both at 4℃ and 0℃ groups. In 0℃ group, the MDA content of erythrocyte and plasma and erythrocyte IF was much lower than that of 4℃ groups (P<0.05),The AOA of plasma, the SOD activities of erythrocyte and plasma, the GSH-Px and CAT activities of blood were all much higher compared with 4℃ group (P<0.05) 21 days after storage. Conclusion: The preserving damages related to the peroxide reaction in the erythrocyte membrane and the obstruction of antioxidation system of blood, which may contribute to erythrocyte injury. Decreasing the preserving temperature can reduce the oxidant injury of the blood to same extent and improve the ED.
