NO在糖尿病大鼠胃排空延迟中作用及机制研究
Effects of NO on delayed gastric empting and its machanism in diabetic rat
【摘要】 为研究 NO在糖尿病胃排空延迟中的作用 ,本文利用 NOS抑制剂 L - NNA和 NO合成的天然底物 L - Arg分别作为处理因素 ,观察在糖尿病不同病程中给予 ,对实验性糖尿病大鼠胃排空的影响。实验结果如下 :早期静脉给予 L- NNA(40 mg/kg) ,对糖尿病大鼠胃排空延迟具有明显的改善作用 ,糖尿病大鼠延迟应用 L- Arg(2 0 0 m g/kg)后 ,也能在一定程度上改善糖尿病大鼠的胃排空延迟 ,血浆 NO含量和胃动素水平较糖尿病模型组亦有改善 (P <0 .0 1) ,这些结果表明 ,糖尿病早期应用 NOS抑制剂能明显改善糖尿病大鼠的胃排空延迟 ,其机制可能是通过降低糖尿病早期过高的 NO含量 ,保护并一定程度上恢复了胰岛 B细胞的内分泌功能 ,从而减轻迷走神经的损伤 ,保护了其调节胃动力的功能
【Abstract】 To study the effect of NO on delayed gastric empting in diabetic rat,we used L-NNA,the NOS inhabitor and L-Arg,the nature substrate of NO synthesis in vivo as treatment measures and observed the effect of L-NNA and L-Arg intravenously infused in different disease phrases in the delayed gastric emptying of the experiment diabetes rats.All the results showed that NOS inhabitor infused in early phrase of diabetes significantly improved the delayed gastric emptying of the experiment diabetes rats,and its mechanism was to reduce the high level of NO in early phrase of diabetes,and to protect the pancreatic insular B cells,and then the damage of vagal nerves was decreased and its ability to adopt the activity of gastric was improved NO nature substrate infused increases the low serum NO level in the late phrase of diabetes and decreases the damage of vagal nerves.
