骨形成蛋白-2基因转染NIH3T3细胞诱导成骨的实验研究

【摘要】 目的 探讨骨形成蛋白 - 2 (bonemorphogeneticprotein - 2 ,BMP - 2 )基因转染NIH3T3细胞体内诱导新骨的能力。方法 用脂质体转染法将BMP - 2基因转染至NIH3T3细胞 ,将转染细胞悬液注入裸鼠股四头肌内 ,第 6周取材 ,观察肌肉组织内诱导新骨生成情况。结果 基因转染实验组裸鼠肌肉组织中有类骨组织形成 ,部分区域伴有钙盐沉着 ,未见明显炎症反应。结论 骨形成蛋白 - 2基因转染NIH3T3细胞在裸鼠肌内具有异位诱导成骨作用

【Abstract】 Objective To investigate the ability in vivo bone formation induced by NIH3T3 cells transfected with bone morphogenetic protein-2(BMP-2) gene. Methods The NIH3T3 cells, which were transfected by liposome mediated delivery of BMP-2 gene, were injected into the quadricep muscles of nude mice. Bone formation of induction in mouse quadriceps 6 weeks after injection was evaluated using histological examination. Results Histological analysis of the group with gene transfection showed that osteoid and some regional calcareous infarct in the mouse muscles were detected, but no inflammatory reaction was observed. Conclusion NIH3T3 cell mediated delivery of BMP-2 gene into mouse muscles could induce ectopic osteogenesis.
