实验性胰腺炎大鼠胰腺总RNA的提取

【摘要】 目的 :探讨提取实验性胰腺炎大鼠胰腺总RNA并尽量减少胰腺RNA降解的方法 ,为胰腺组织mRNA的检测提供前提条件 .方法 :健康SD大鼠 5 4只 ,随机分为假手术组 (A)、损伤组 (B)、动脉给药组(C)和静脉给药组 (D) ,RNA提取采用Chomczynski酸性一步法 .结果 :RNA琼脂糖凝胶电泳显示RNA没有明显降解 .结论 :胰腺组织RNA含量丰富 ,极易降解RNA ,操作速度、RNA提取试剂盒的质量和防止RNA污染的综合措施是成功制备高质量RNA的关键

【Abstract】 Objective: To explore the methods of preparating total RNA of pancreas in rats and reducing the degradation of RNA in order to provide preconditions as the base of analysis of mRNA in pancreas. Methods: 54 rats were divided into 4 groups randomly, including sham operation group, artificial AP group, dopamine treating group by arterial injection and dopamine treating group by tail vein injection. RNA preparation was based on Chomczynskis one step method. Results: RNA was not obviously degradated as the result of RNA agrose electrophoresis. Conclusion: The pancreas in rat is abound in Rnases, which makes RNA in pancreas stay in high risk of degradation. The important factors for gaining high quanlity of RNA from pancreas are processing quickness of preparation RNA and the quality of reagent.
