HPLC法测定环磷酰胺血药浓度
Determination of cyclophosphamide in human plasma by HPLC
【摘要】 目的 :建立以甲醇 -水为流动相 ,用HPLC法测定环磷酰胺血药浓度的方法。方法 :用氯仿萃取血清中的环磷酰胺 ,浓集后进样测定。在室温条件下 ,使用ELITEC18色谱柱 (5 μm) ,以甲醇 -水 (1∶1)为流动相 (流速1.0mL/min) ,以咖啡因 (0 .1g/L)为内标 ,检测波长 2 0 0nm。结果 :本方法线性范围 10mg/L~ 2 0 0mg/L ,检测限5 .5mg/L ,相关系数 0 .9994 ,回收率 97%~ 10 5 %。结论 :方法灵敏、简便、准确 ,适用于对环磷酰胺进行治疗药物监测的需要
【Abstract】 Objective: To develop a method of determinining the concentration of cyclophosphamide in human plasma by HPLC. Methods: Extract cyclophosphamide in human plasma with CHCl3, determine its concentration after concentrating by HPLC. Results: The linear range of this method is 10~200mg/L, the correlation coefficient is 0.9994 ( r = 0.9994, n = 7 ), the limit of detection is 5.5mg/L. Conclusion: This method is quite sensitive?simple and accurate, it is suitable to be used in determining the concentration of cyclophosphamide in human plasma.
