红景天水煎剂对心肌缺血再灌注损伤的保护作用

【摘要】 目的 观察红景天对心肌缺血再灌注损伤的保护作用 ,并初探机制。方法 SD大鼠 30只 ,随机等分为三组 :假手术组 (A组 ) (生理盐水 2ml/d)、I/R模型组 (B组 ) (生理盐水 2ml/d)、I/R模型 +红景天组 (C组 ) (5 g/kg/d)。连续灌胃 5d后 ,A组完成模型全部操作 ,只穿线不结扎冠脉 ;B、C组完成模型全部操作 ,穿线并结扎冠脉 ,缺血 6 0min ,再灌注 6 0min后处死。各组均取全血离心检测血中TNF -α、IL - 6、ET ;透射电镜观察心肌细胞超微结构。结果 C组与B组相比 :显著降低血中炎性细胞因子TNF -α、IL - 6、ET水平 ;心肌细胞超微结构的变化情况 :B组与A组比较其超微结构发生异常改变 ;C组与B组比较 ,心肌细胞超微结构的异常改变明显减轻。结论 红景天对心肌缺血再灌注损伤具有保护作用 ,这种保护作用部分与其降低细胞因子及ET水平有关

【Abstract】 Objective To observe the protective effects of Hongjingtian on ischemia and reperfusion (I/R) Injury in rat heart and the possible action mechanism .Methods 30 male Spraegu-Dawley rats were randomly divided into three groups( n=10 in each): sham operated group ( group A, normal saline 2ml/d), I/R group( group B, normal saline 2ml/d), Hongjingtian-treated I/R group( group C, 5g/kg/d). After five days of pretreatment, the left anterior descending coronary arteries(LAD) of the ten rats in group A were false ligated. In group B and C , the LADs were ligated and obturated for 60 minutes ,and then reperfused for 60 minutes. The whole bloods of all rats in each groups were collected and then centrifuged to determine the content of serumal TNF-α,IL-6,ET. The ultrastructures of injured myocardial cells by electron microscope were also observed.Results The concentration of TNF-α,IL-6,ET were remarkably decreased and there was both obvious significance in the treatment group compared with model group. The results of electron microscopy also showed the same direction.Conclusions The results demonstrate that hongjingtian provides remarkable protective effects in a rat model of ischemia reperfusion and the protective effects of Hongjingtian is partially due to reducing the concentration of TNF-α,IL-6,ET.
