HPLC测定人血浆中不对称性二甲精氨酸浓度

【摘要】 目的 建立一种测定人血浆中不对称性二甲精氨酸 (ADMA)浓度的高效液相色谱法 (HPLC)。方法 采用HypersilC18分析柱 ,以磷酸盐缓冲液 :甲醇 ( 5 8:4 2 )为流动相 ,流速 1.0mL/min ,荧光检测 ,激发波长 (λex) 338nm ,发射波长 (λem) 4 2 5nm ,以邻苯二甲醛 (OPA)为衍生剂进行柱前衍生 ,室温下检测。结果 ADMA在 5 0~ 6 0 0ng/mL范围内有良好线性关系 ,最小检出量为 5 0ng/mL ,回收率为 96 .37%~ 10 2 .72 % ,日内、日间平均变异系数分别为 4 .6 7%和 5 .5 2 %。结论 本方法准确、简便、快速 ,适用于临床检测

【Abstract】 Objective To evaluate the determination of asymmetric dimethylarginine(AMDA)in human plasma with highperformance liquid chromatography(HPLC).Methods Human ADMA was analysed by HPLC with a column of Hypersil C18,mobile phase of phosphate buffer methanol(58:42 V/V),at a flow rate of 1.0 mL/min,at room temperation and detection fluorescence with excitation wavelength(λex) at 338nm,emission wavelength(λem) at 425nm,derivative before column using o phthaldialdehyde mercaptoethanol(OPA).Results the linear range was 50ng/mL to 1600ng/mL.The minimum limit of detection was 50ng/mL.The recoveries were 96.37%~102.72%.The coefficient of variation for intra and inter were 4.67% and 5.52%.Conclusion The method is very accuracy, simple and quick, which is suited for usage in clinical diagnosis.
