甲基莲心碱对移植至裸鼠的增生性瘢痕胶原和酸性粘多糖合成的影响
Influences of Neferine on Synthesis of Collagen and Acidic Mucopolysaccharide in Hypertrophic Scars in Nude Mice
【摘要】 目的 探讨甲基莲心碱 (Neferine ,Nef)对移植至裸鼠的人增生性瘢痕的作用。方法 建立人增生性瘢痕裸鼠动物模型 ,将瘢痕组织块植入 18只裸鼠皮下 ,术后第 2 4d组织成活后 ,随机分为 3组 :①Nef治疗组 (A组 ) ;②稀盐酸对照组 (Nef溶媒 0 .0 3MHCl∶NS =1∶9) (B组 ) ;③空白对照组 (C组 )。A、B组每只每 2d分别局部注射Nef10 0 μl(1.2mg/ml)和稀盐酸 10 0 μl。两周后 ,用光、电镜观察组织和细胞形态学变化 ,测量植入物的体积 ,用全自动图像分析仪测量组织中胶原和酸性粘多糖含量。结果 A组植入物体积缩小 ,与B、C组比较 ,差异有非常显著性意义 (P <0 .0 0 1) ;胶原排列较整齐 ;Ⅰ型胶原减少 ,Ⅲ型胶原增加 ;胶原和酸性粘多糖含量显著降低 (P <0 .0 0 1)。结论 Nef对移植至裸鼠的人增生性瘢痕有减小体积 ,改变Ⅰ、Ⅲ型胶原成分 ,降低胶原和酸性粘多糖含量的作用。表明Nef对移植至裸鼠的增生性瘢痕有一定的治疗作用
【Abstract】 Objective To investigate the effects of nferine(Nef)on human hypertrophic scars in nude mice.Methods The animal model was established in nude mice with human hypertrophic scars as following:scars were implanted into the subcutaneous tissues of 18 nude mice that were randomly divided into 3 groups(group A,group B and group C)at 24 d postoperatively after survived.Every mouse of group A and group B was injected locally with 100μl Nef( 1.2 mg/ml)and hydrochloric acid HCl(dissolvent of Nef 0.03M HCl:normal saline =1:9)for 2 weeks,respectively.Group C was controls.The morphological changes of the harvested tissues were observed under light microscope and electron microscopy,and compared with the originals.The contents of collagen and acidic mucopolysaccharide in the tissues were measured respectively by means of automatic image analysis system.Results The average volume of the tissues in group A shrunk[A=(0.201 3±0.028 2)cm 3,B=(0.311 8±0.037 8) cm 3,C=(0.314 3±0.085) cm 3].The results showed significant difference between group A and B and between group A and C( P <0.001).In group A, the arrangement of collagen fibers was in better order than that of the control group.Collagen type Ⅲ was increased,while collagen typeⅠwas reduced.The contents of collagen and acidic mucopolysaccharide were reduced(collagen:A=0.495 2±0.029 3,B=0.791 3±0.022 5,C=0.791 0±0.029 3;acidic mucopolysaccharide:A=0.270 8±0.015 7,B= 0.410 4±0.050 3,C=0.409 1±0.053 1)( P <0.001).Conclusion Local injection of Nef into the nude mice with scars could help reduce the volume of the scars,change the contents of type Ⅰand Ⅲ collagen,decrease the contents of collagen and acidic mucopolysaccharide.
