川芎嗪对大鼠缺血心肌Bcl-2及Bax蛋白表达的影响

【摘要】 目的 :研究心肌缺血时凋亡相关基因Bcl 2和Bax蛋白表达水平的变化及川芎嗪对它们的影响。方法 :大鼠随机分对照组、缺血组和川芎嗪保护组。采用皮下注射异丙肾上腺素 (ISP ,5mg/kg) ,2 4h后再注射同剂量一次 ,造成心肌缺血损伤模型。采用免疫组化法检测Bcl 2和Bax蛋白水平的变化。结果 :缺血组Bcl 2和Bax蛋白表达均高于正常组 (P <0 .0 5和P <0 .0 1) ,川芎嗪可显著提高缺血心肌组织Bcl 2的表达 (P <0 .0 1) ,对Bax的表达影响不大 (P >0 .0 5 )。结论 :在异丙肾上腺素所致的心肌缺血性损伤中 ,凋亡相关基因表达异常 ,川芎嗪对该模型大鼠心肌Bcl 2基因的异常表达有一定的调控作用

【Abstract】 Objective:To study the relationship between the protein expressions of some apoptosis related genes and myocardial ischemical changes,and to observe the effects of ligustrazine on expressions of Bcl 2 and Bax proteins.Methods:The rats were randomly divided into the control, model,and ligustrazine treated groups.The model of myocardial ischemia was induced by subcutaneous injection of isoproterenol (ISP,5 mg/kg)into rats and repeated by the same dose 24 hours later.The changes of protein expressions of Bcl 2 and Bax were detected by immunohistochemical staining.Results:In the model group,as compared with those of control,the protein expressions of Bcl 2 and Bax were increased( P <0.05 and P <0.01). Ligustrazine enhanced the protein expression of Bcl 2,but did not significantly Bax expression( P >0.05).Conclusion:Subcutaneous injection of ISP could induce protein expression of apoptosis related genes in rat myocardium,and expression of Bcl 2 is influenced by ligustrazine.
