6-(3-甲基-2-羟基-3-烯-1-丁基)-3,4-二甲氧基-2,5-二苄氧基甲苯的合成
Synthesis of 6-(2-Hydroxy-3-methylbut-3-enyl)-3,4-dimethoxy-2,5-dibenzyloxytoluene
【摘要】 以辅酶Q0为原料,通过三步主要反应,合成了辅酶Q10的重要中间体6 (3 甲基 2 羟基 3 烯 1 丁基) 3,4 二甲氧基 2,5 二苄氧基甲苯。第1步Friedel Crafts烷基化反应以三氟化硼乙醚络合物为催化剂,收率69 9%。环氧化反应用过氧乙酸〔w(CH3CO3H)=40%〕作氧化剂,得到95 3%的收率。最后采用异丙醇铝进行选择性开环得目标产物(Ⅲ),收率89 9%。总收率达到60%(以辅酶Q0计)。
【Abstract】 6-(2-Hydroxy-3-methylbut-3-enyl)-3,4-dimethoxy-2,5-dibenzyloxytoluene,an important intermediate of coenzyme Q10,was synthesized through three-step process employing coenzyme Q0 as the starting material.The first step,involving Friedel-Crafts alkylation reaction with boron trifloride etherate as the catalyst,gave 69.9% yield.In epoxidation reaction with peracetic acid 〔w(CH3CO3H)=40%〕 as the oxidant,95.3% yield was realized.In the last step,the selective ring-opening reaction utilizing aluminum triisopropoxide,the allylic alcohol target product was obtained in 89.9% yield.The overall yield reached 60%(based on coenzyme Q0).
