农杆菌介导的玉米大斑病菌转化(英文)

【摘要】 采用玉米大斑病菌Exserohilum turcicum的无性菌丝作受体,建立了农杆菌Agrobacterium tumefaciens介导的转化方法。采用的载体为 pUR5750, 携有来源于质粒pAN71的潮霉素抗性选择标记。PCR分析表明T-DNA 插入到E. turcicum基因组中。

【Abstract】 A transformation technique for Exserohilum turcicum mediated by Agrobacterium tumefaciens using vegetative mycelium as recipient was developed and procedure for the enhancement of transformation efficiency was optimized. The binary vector used in transformation was pUR5750, which contains selection marker gene conferring hygromycin-resistance under control of the Aspergillus nidulans gpd promoter and trpC terminator between the left border and right border repeats of the binary vector pBIN19. PCR analysis showed that T-DNA was integrated into the genome of E. turcicum.
