胶质瘤对血脑屏障内皮细胞间连接的影响
Influence of glioma cells on intercellular junctions of blood-brain barrier
【摘要】 目的:探索胶质瘤细胞对血脑屏障细胞间连接的作用及其分子机制。方法:利用内皮细胞与胶质细胞共培养建立体外血脑屏障模型,采用银染的方法探索胶质瘤细胞对其中内皮细胞间连接的作用并运用半定量RT-PCR的方法分析胶质瘤细胞作用后内皮细胞紧密连接蛋白1(ZO-1)的表达变化。结果:在胶质瘤细胞的直接作用过程中,血脑屏障细胞间连接的完整性受到明显破坏,内皮细胞ZO-1表达水平降低;在其间接作用过程中,两者变化不明显。结论:胶质瘤细胞的直接浸润可以破坏血脑屏障内皮细胞间连接,而其间接作用则不能完全破坏。胶质瘤细胞对内皮细胞ZO-1表达的影响是胶质瘤发生、发展过程中血脑屏障内皮细胞间连接变化的重要分子基础。
【Abstract】 Objective :To investigate the influence of glioma cells on intercellular junctions of blood-brain barrier and its molecular mechanism. Methods : A blood brain barrier model was established by coculture of ECV304 and astrocytes in vitro. Morphological changes of intercellular junctions in the blood brain barrier was determined by silver staining. The expression level of zonula occluden 1(ZO-1) was analyzed with semiquatitative RT-PCR. Results : The integrity of the BBB intercellular junctions were destroyed by the direct influence of glioma cells, but the indirect influence of them could not change the integrity. The expression level of ZO-1 was elevated by the direct effects but not by the indirect influence of the glioma cells. Conclusion : Intercellular junctions of BBB can be disrupted by the direct influence of glioma cells. Changes of ZO-1 expression level may be one of the most important molecular bases of this direct influence.
