大鼠周围神经损伤后肌肉组织乙酰胆碱酯酶的变化
Changes of acetylcholinesteras level in muscle after rat peripheral nerve injury
【摘要】 目的:探讨周围神经损伤后,在不同时间点受神经支配的肌肉组织中乙酰胆碱酯酶(AChE)的变化。方法:建立大鼠外周神经损伤的动物模型;制备特异识别神经系统AChE的单克隆抗体,以该抗体作为一抗,酶标羊抗鼠IgG作为二抗建立酶联免疫检测方法;测定正常对照组、坐骨神经神经损伤后0.5 h、3 h、12 h组,坐骨神经断点处、胫神经及腓总神经所支配的肌肉组织AChE的含量变化。结果:坐骨神经损伤0.5 h后,断点处肌肉组织中AChE含量最高,随时间延长逐渐降低;胫神经、腓总神经支配的肌肉组织中AChE含量0.5 h时最低,3 h最高,12 h时降低。结论:坐骨神经损伤后,断点处及其远端神经支配的肌肉组织中AChE的变化呈时间依赖性,但变化不一。
【Abstract】 Objective:To explore the change of acetylcholinesterase(AChE)in nervous innervated muscle tissues after the nerves were injured at different time points. Methods:The rat peripheral neve injury model was established,and the monoclonal antibody to nervous AChE were prepared and used as first antibody, selected antibody IgG-HRP from sheep to mouse as second antibody,ELISA was carried out to detect the AChE level of muscle in different group. Result:The AChE level of nerve innervated muscle were the highest after sciatic nerve injury 0.5 h after injury, then it descended gradually as time went by. The AChE level of tibial nerve and common peroneual nerve innervated muscle were the lowest 0.5 h after injury the change were the highest 3 h after injury, All of them were declined 12 h after injury Conclusion: The change of AChE level in different muscle are different depending on the time after nerve injured at different time points.
