【摘要】 目的 :研究Aβ对神经元膜流动性的影响 ,探索Aβ的神经毒性机制。 方法 :制备急性分离的新生SD大鼠海马和皮层神经元细胞悬液 ,采用显微准弹性激光散射技术和荧光偏振技术。结果 :采用显微准弹性激光散射技术发现Aβ2 5~ 3 5作用 5min和 35min后均降低海马和皮层神经元ACF的衰减速率及频移线宽Γ ,而采用荧光偏振技术发现Aβ2 5~ 3 5作用 5min和 35min后虽然有升高海马和皮层神经元荧光各向异性的趋势 ,但无统计学意义。结论 :提示Aβ可降低神经元膜流动性 ,显微准弹性激光散射技术比荧光偏振技术更敏感。

【Abstract】 Objective:The effect of Aβ on neuronal membrane fluidity was studied to explore the neurotoxic mechanism of Aβ on neurons.Methods:Freshly dissociated hippocampal and cortical neuron suspension of newborn SD rats were prepared.Two methods(MQLS and FPT) were employed to measure the membrane fluidity.In the microscope quasi-elastic laser light scattering(MQLS) technique,the neurons’ autocorrelation function(ACF) of the scattering light intensity was analyzed.The frequency shift line width Γ was fitted by ACF.The Γ could symbolize the cell membrane fluidity.In the fluorescence polarization technique,DPH was used and its anisotropy could denote the cell membrane fluidity.Results:The ACF attenuate rate and the frequency shift line width Γ of both hippocampal and cortical neurons decreased significantly by Aβ acting on neurons for 5 min and 35 min.The anisotropy of DPH increased slightly but insignificantly after the application of Aβ for 5 min and 35 min.Conclusion:The data indicate that Aβ could make neuronal membrane fluidity decrease,resulting in neurotoxicity and MQLS technique is more sensitive than fluorescence polarization technique.
