MMP-9与人黑色素瘤细胞生长相关性的研究
A STUDY ON THE CORRELATION BETWEEN MATRIX METALLOPROTEINASE 9(MMP-9)AND THE GROWTH PHENOTYPE IN HUMAN MELANOMA CELLS
【摘要】 目的 利用多种方法检测金属蛋白酶MMP - 9基因与人黑色素瘤细胞生长的相关性。方法 利用细胞记数、MTT、3H -thymidine掺入实验等方法检测分别转染正义和反义MMP - 9CDNA后人黑色素瘤细胞生长速度的改变。结果 转染反义基因后WM 4 5 1细胞生长速度受到明显抑制 (P <0 .0 5 ) ;转染正义基因后WM 35细胞生长速度受到明显促进 (P <0 .0 5 )。而运用四环素可以抑制四环素负调控逆转录病毒载体上的外源基因的表达 ,其结果与对照细胞相似。结论 反义MMP - 9基因使人黑色素瘤细胞的生长能力受到一定程度的抑制 ,正义MMP - 9基因使人黑色素细胞的生长能力受到一定程度的促进。说明MMP - 9在人黑色素瘤细胞生长过程中起重要作用。同时 ,四环素负调控逆转录病毒载体可以调控外源基因的表达
【Abstract】 Objective To investigate the correlation between MMP-9 and tumor growth.Methods One self-contained tetracycline-regulated retroviral vector containing antisense cDNA of MMP-9 was constructed and transfected into a metastatic human melanoma cell line WM451, which grows fast and has a high MMP-9 expression level; at the same time sense cDNA of MMP-9 was constructed and transfected into a metastatic human melanoma cell line WM35, which grows slowly and has a low MMP-9 expression level. Tests such as growth rate, MTT method, 3 H-thymidine incorporation were used to analyse the growth phenotype of tumor cells before and after gene transfection. Results After transfection of antisense MMP-9, the capacity of in vitro growth was significantly suppressed(P<0.05); after transfection of sense MMP-9, the capacity of in vitro growth was significantly promoted (P<0.05). Conclusion The decrease of MMP-9 expression in human melanoma cell line by MMP-9 antisense RNA could inhibit its growth phenotype and the increase of MMP-9 expression in human melanoma cell line by MMP-9 sense RNA could hasten its growth phenotype. All these demonstrated that MMP-9 played an important role in tumor growth.
