抗HIV植物蛋白MAP30等药物的体外抗单纯疱疹病毒作用
Anti-herpes simple viruses effects of anti-HIV plant protein MAP30 and other three drugs in vitro
【摘要】 目的 :研究抗人类免疫缺陷病毒 (HIV)植物蛋白MAP30、无环鸟苷 (ACV)、α 干扰素 (IFN α)及叠氮胸苷(AZT)等药物对单纯疱疹病毒 (HSV)的体外抑制作用。 方法 :以Vero细胞为靶细胞 ,观察药物作用 4 8h后对HSV致细胞病变的抑制作用 ,并采用ELISA法测定药物对培养上清HSV 1和HSV 2抗原分泌的抑制作用。 结果 :MAP30和ACV均可使HSV的致细胞病变效应减轻。MAP30对HSV 1和HSV 2抗原分泌的 5 0 %抑制浓度 (IC50 )分别是 0 .5 μmol L和 0 .4 μmol L ,ACV分别是 2 .8μmol L和 2 .2 μmol L ,而IFN α和AZT对HSV无明显抑制作用。 结论 :MAP30除具有抗HIV活性外 ,还具有抗HSV的作用。
【Abstract】 Objectives: To examine the effects of anti HIV plant protein MAP30,ACV(acyclovir),IFN α (α interferon),AZT (3′ azido 2′,3′ dideoxythymidine) on the infection and replication of herpes simple viruses(HSV). Methods: Vero cultured in the presence of tenfold dilutions of these drugs were exposed to HSV 1 and HSV 2, and viral yield was measured at 48 hours by ELISA.The inhibitive effect of these drugs on HSV induced cytopathicity was also observed. Results: The 50% inhibition concentrations (IC 50 ) were 0.5 μmol/L for HSV 1,and 0.4 μmol/L for HSV 2 for MAP30, respectively. In comparison, the IC 50 for ACV, a commonly used anti HSV, was 2.8 μmol/L and 2.2 μmol/L for HSV 1 and HSV 2, respectively.Cytopathic effect of HSV was also inhibited by MAP30 and ACV. IFN α and AZT showed little effect on HSV. Conclusions: These results suggest that MAP30, previously showed to be active against HIV, may be useful for the therapy of HSV infections.
