肠上皮细胞凋亡与肠缺血损伤及再生修复的实验研究

【摘要】 目的 探讨小肠黏膜缺血损伤及再生修复的特征及规律 ,以及与肠上皮细胞凋亡的关系。方法 应用小肠缺血再灌注模型 ,检测小肠缺血及再生修复过程中肠黏膜结构、黏膜隐窝上皮细胞分裂活性及黏膜上皮细胞凋亡的改变。结果 肠缺血再灌注 1h ,肠黏膜损害加重 ,黏膜上皮细胞凋亡明显增加 ,黏膜隐窝上皮细胞分裂活性降低 ;肠黏膜再生 1d、 3d组肠黏膜上皮细胞凋亡明显减少 ,黏膜隐窝上皮细胞分裂活性增加 ,并逐渐恢复至正常水平 ,肠黏膜结构逐渐恢复至正常。结论 肠上皮细胞凋亡在肠缺血再灌注损伤及肠再生修复过程中起重要作用

【Abstract】 Objective To investigate the relationship between the apoptosis of intestinal epithelium and characteristics of intestinal mucosal ischemia-reperfusion damage and repair after injury in rats.Methods An experimental model of intestinal ischemia-reperfusion in rats was established.Intestinal mucosal morphology and epithelial apoptotic and mitotic activities were assessed 1,24,72 hours after a 60-minute vascular occlusion of a 15-cm segment of midintestine in rats.Results Progressively severe mucosal injury,dramatically increased epithelial apoptosis and decreased epithelial mitotic activity occurred one hour after reperfusion of the ischemic intestine.At the 24 hours stage,the recovery of mucosal injury and epithelial mitotic activities and attenuation of epithelial apoptosis were observed.After 72 hours,intestinal mucosal morphology and epithelial apoptotic and mitotic activity returned to normal gradually.Conclusion Apoptosis of intestinal epithelium plays an important role in intestinal mucosal ischemia-reperfusion damage and repair.
