小鼠对HPV16L1-E7重组腺病毒和重组质粒的免疫应答
Immune response induced by recombinant HPV16 L1-E7 in adeno-virus vector and in plasmid
【摘要】 对比研究人乳头瘤病毒 16型L1 E7重组腺病毒 (rAd5HPV16L1 E7)和重组质粒经不同途径免疫小鼠所产生的免疫效应。将rAd5HPV16L1 E7病毒在 2 93细胞中扩增 ,并制备rAd5HPV16L1 E7重组质粒 ,分别以肌肉注射、滴鼻途径免疫小鼠 ,采用ELISA法测其血清IgG抗体水平 ;制备脾细胞悬液 ,加入到 96孔无菌培养板中 ,分别加入rAd5HPV16L1 E7特异性蛋白和 10 %FCSRPMI16 4 0 ,加入3 H TdR 1μCi/孔 ,做T 细胞增殖实验 ;取经过培养 5 6h的培养液进行IFN γ的测定。不同疫苗和不同途径免疫的BALB/c小鼠血清IgG抗体水平均高于对照组小鼠 (P <0 0 5 ) ,IFN γ和T 细胞增殖亦高于对照组 (P <0 0 5 ) ,且各实验组中病毒特异性蛋白刺激组均高于非刺激组 (P <0 0 5 )。提示 :rAd5HPV16L1 E7重组质粒和rAd5HPV16L1 E7重组活病毒一样既能刺激T 细胞产生细胞免疫应答 ,又能刺激B 细胞产生体液免疫应答 ,说明其具有很好的免疫原性 ,是一种很有发展前景的防治疫苗
【Abstract】 To study the immune response of BALB/c mice immunized by HPV 16 L1 E7 recombinant adeno virus type 5 (rAd 5HPV 16 L1 E7) and HPV 16 L1 E7 instetel plasmid. rAd 5HPV 16 L1 E7 virus was propagated in 293 cell line and then rAd 5HPV 16 L1 E7 plasmid was prepared .The BALB/c mice were immunized with two vaccines by intramusclar injection or nasal drip. The serum IgG level of these mice was tested. The spleen cells (were) cultured. T cell proliferation test was done and IFN γ in supernatant cultured of mice spleen cell culture was tested. The level of serum IgG in the immunized BALB/c mice was higher than that in the control mice( P <0.05). The data of IFN γ level and T cell proliferation indicated that the immunized mice by different immuno paths was higher than the controls( P <0.05). rAd 5HPV 16 L1 E7 plasmid as rAd 5HPV 16 L1 E7 virus, also stimulated the mice to activate not only T cell immunoresponse but also humoral immunoresponse. It suggested both of them could be candidates as prophylactic and therapeutic vaccines.
