重组耻垢分枝杆菌-Sj26GST疫苗的免疫保护作用研究

【摘要】 目的 初步研究重组耻垢分枝杆菌疫苗 r Mc- Sj2 6 GST (recom binant M.smegmatism c2 1 5 5 - Sj2 6 GST)对小鼠的免疫保护作用。方法 用耻垢分枝杆菌重组疫苗 r Mc- Sj2 6 GST免疫雄性 BAL B/ c小鼠 ,检测小鼠淋巴细胞刺激指数 (SI) ,腹腔巨噬细胞培养上清释放 NO量 ,小鼠血清IFN- γ、IL- 2的含量 ,并计数小鼠肝、肺活细菌数。结果 小鼠脾淋巴细胞刺激指数 (SI)为 2 .6 4±1 .37,与 Mc组 (载体组 ) 1 .2 8± 0 .4 1相比 ,差异具有显著性 ;小鼠血清 IFN- γ为 1 96 .4 3pg· ml- 1 ,与 Mc组 1 1 2 .5 7pg· ml- 1相比 ,差异有显著性 ,较对照组高 4 1 % ;小鼠血清中 IL- 2的浓度较对照组高。经 r Mc- Sj2 6 GST疫苗免疫的小鼠受结核杆菌攻击后其肺、肝脏结核杆菌数较对照组少。结论 耻垢分枝杆菌重组疫苗 r Mc- Sj2 6 GST增强了小鼠细胞免疫功能 ,并使小鼠能抵抗结核杆菌的攻击

【Abstract】 Objective To study the protective immune function of recombinant M.smegmatis bearing schistosoma japonicum 26kD antigen gene. Methods BALB/c male mice were immunized with recombinant M.smegmatis (rMc Sj26GST) vaccine. Detected the stimulating index (SI) of splenic lymphocyte transformation and the amounts of NO in supernatant released from the intraperitoneal macrophage. The levels of IFN γ and IL 2 in the serum were detected. Live bacterial CFU in livers and lungs were calculated. Results The SI of the experimental group was 2.64± 1.37 , which was significantly higher than that of vector group (1.28±0.41). The serum IFN γ level of the experimental group was 196.43±66.12 pg·ml -1 , which was significantly higher than that of vector group (112.57±44.78 pg·ml -1 ). Compared with control group (139.29±65.73 pg·ml -1 ), the serum IFN γ level of experimental group increased by 41%. The serum IL 2 level of experimental group was significantly higher than that of control group. Live bacterial CFU in livers and lungs were lower than that of control group. Conclusions The vaccine could effectively stimulate host cells generate IL 2 and IFN γ, and induce extremely strong protective immunity against subsequent challenge with M.tuberculosis H37Ra.
