离体小麦胚胎对6种植物激素的生长反应
Growth Reaction of Wheat Embryos Cultured in Vitro to 6 Kinds of Plant Hormone
【摘要】 通过对38种培养基的筛选试验,结果指出,小麦近成熟胚和成熟胚在试管中生长,基本培养基仍以N6为优。附加0.1~2.0的IBA(单位为mg/L,植物激素下同)和不大于0.5的IAA可使胚胎快速萌发并生长成正常植株;若再添加0.5及以上的KT或6-BA,对胚胎萌发和正常生长不利。单因子的6-BA附加0.1浓度即排斥根系下扎入培养基。NAA和2,4-D一样,对小麦胚胎有脱分化作用,单因子0.5的NAA和0.1的2,4-D即可使根、芽生长受阻,进入脱分化,并随浓度提高,脱分化作用增强。加入0.3%的活性炭可明显减弱6-BA、NAA和2,4-D的脱分化作用,使植株正常生长。
【Abstract】 To culture immature and mature embryos of wheat in vitro, the N6 basic medium is batter than others. Adding 0. 1~2.0mg/1 IBA or<0. 5mg/1 IAA into the medium,the embryos germinate root and leaf normal. If adding KT or 6-BA again at the concentration of 0. 5 mg/l,the embryos growth become badly. Roots grow abnormally and do not take themselves into medium in which contains 0. 5 mg/1 6-BA. NAA,as same as 2,4-D,can make wheat embryos un-differentiation at and over the concentration of 0. 5 mg/1. but for the 2,4-D,it needs only at 0. 1 mg/1. The active carbon at 0. 3% concentration could weaken the un-differentiation of 6-BA,NAA and 2,4-D,and make the plantlet grow normal.
