Ce(Ⅲ)-ALC络合物与鱼精子DNA相互作用的电化学与光谱研究
Studies of Ce(Ⅲ)-ALC interacting with herring sperm DNA by electrochemical and spectrophotometric methods
【摘要】 用电化学方法和荧光光谱法,UV光谱法对Ce( )-ALC络合物与鱼精子DNA的相互作用进行了研究,发现在在pH为4.3的六次甲基四胺((CH2)6N4)缓冲溶液中,Ce( )-ALC络合物与鱼精子DNA(hsDNA)以嵌入方式生成一种电化学非活性络合物Ce( )-ALC-DNA,导致Ce( )-ALC的峰电流降低,峰电流在一定范围内与DNA的浓度成线性关系,线性范围为1~10μg/mL,相关系数为0.997,检测限为0.1μg/mL,因此该法可以用于DNA的测定.
【Abstract】 In this paper, the interaction of Ce(Ⅲ)ALC and herring sperm DNA(hsDNA) is studied by electrochemical, fluorimetric and UVspectrophotometric methods. Ce(Ⅲ)ALC complex can react with hsDNA to produce an electrochemically nonactive supermolecular complex Ce(Ⅲ)ALCDNA in the mode of intercalation in the buffer solution of (CH2)6N4(pH=4.3), which results in the decrease of the peak current of Ce(Ⅲ)ALC. The decrease of the peak current is proportional to DNA concentration from 1 to 10 μg/mL, the limit of detection is 0.1 μg/mL, and the correlation coefficient is 0.997, this method can be applied to determine DNA concentration.
