气单胞菌几丁质酶的性质与发酵条件
Analysis of Aeromonas sp. CJ-5 producing chitinase
【摘要】 从浙江省土样中分离出一株产几丁质酶的好氧菌,经鉴定该菌属于气单胞菌属(Aeromonassp.),命名为CJ-5菌株.经研究发现,CJ-5菌产生的几丁质酶是一种诱导、分泌型酶,其产酶的最佳发酵条件为:起始pH7.0,温度为30℃,几丁质底物质量浓度为2%~2.5%,培养基装量为三角烧瓶总体积的20%.在3L发酵罐中进行了扩大培养,发酵液的最高几丁质酶活力为0.41U·mL-1.该酶的酶促反应最适温度为36℃,最适pH为7.0.在进一步的研究中,还发现CJ-5菌株是一株产双酶的菌株,既产几丁质酶,又产壳聚糖酶.
【Abstract】 An aerobic strain capable of producing chitinase was isolated from soil samples in Zhejiang Province. It was identified to belong to Aeromonas sp. and named CJ-5 strain. The chitinase produced by CJ-5 strain is inducible and secretory. The optimal fermentation condition for CJ-5 strain producing chitinase is to incubateit in flask at 30 ℃, with 20 % volume medium containing 2 %-2.5 % chitin, initial pH7.0. The chitinase activity of CJ-5 strain reaches 0.41 U·mL-1 when it is incubated in 3 L fermentor. The optimum pH and temperature for hydrolyzing chitin are 7.0 and 36 ℃, respectively. It was found that CJ-5 strain produces not only chitinase, but also chitosanase.
